      "While we know that the single dose can protect against disease" seems like a strong statement - these were post hoc analyses, right? Your piece doesn't help any reader understand why post hoc analyses are problematic.

      Weak yes - which is why we need the study. However, the data we DO have shows a 92% efficacy among people without a second dose. I don't see how this is so weak we should not seriously consider it. 2/983 vs 28/1059. Durability unknown - some ppl go out to 108 days.

      A possible flaw with the single-dose idea: What would the comparison group be for these trials? If it's unethical to deny placebo-takers the vaccine, wouldn't it also be unethical to deny two doses to a single-dose taker if the latter provides better protection? 1/2

      One option would be to offer vax+vax/placebo trial to groups not getting any vaccine right now. Given the shortage, there is no way avoiding denying something to large numbers of people; and if we don't try but later find single-dose holds up, we've denied maybe a billion people.

      Plus, there will be people who miss the second dose anyway, and there will need to be an effort to track them down. Just saying "please don't miss your dose" won't work; these things need resources. With a trial, we can make a smarter decision on that.

          Hmm, a retro trial with people who miss their second dose would be an interesting way to do it. Randomization and measuring immune responses would be tricky....

          I still feel like the immunology in the Moderna trial is telling us that the single dose may not work. Also plenty of childhood vaccine schedules require 2 doses w/o creating many issues. But I get where you’re coming from and it’s interesting pitch.
