Plus, the US is planning to hold off vaccinating half the people *right now*, keeping the second dose for them in the freezer merely in case of production hiccup. At a minimum, can we discuss vaccinating as many a possible now, given a few days off doesn't seems a terrible risk?

How? We will have vaccinated twice as many and the production is already scheduled for the booster. Let's assume there's a hiccup and a delay of a few days. The data above very very strongly suggests it wouldn't even be detectable. (Immunity *may* wane long term, not in days).

Once again, @ScottGottliebMD and I are on the exact same page about this. It doesn't make sense, in the worse phase of the pandemic to date, to hold back so many doses of the vaccine when we know the first dose can have an effect. Good explanation here. https://twitter.com/SquawkCNBC/status/1337396511957716996 …

also, how about some single dose trial data first, so we really know what we are doing? #covid19 #vaccine

in an ideal world, with lots of time, we would. this gets at the necessity of 100% perfect information.

But @walidgellad it's a leap from what we have from studies to date to extrapolating single dose efficacy. What is duration of single dose response? Will we see infections as immunity wanes? I think focus should be on efficient & widespread production & administation of 2 doses.

no one knows answer to your questions. my argument has been that the probability most people will miss 2nd dose or be very delayed is small, and in 3 months the world will be very different.

Yeah, I get your argument. As you point out, many have advanced it. We can't know now what approach would yield greater good. It's not waiting for "perfect" data on single dose, it's waiting for any real data in absence of later giving a second dose as in trials to date.

Agree, very encouraging, certainly out to d 28, & then beyond w 2nd dose. We don't really know w single dose on duration of effect & impact on transmission. Absent data, it's what your threshold is for leap of faith on single dose. Thx for your great work, too. #covid19 #vaccine

Thanks! I would absolutely like more data as well though I guess I’d phrase it a little differently. Immunity is not going to disappear on day 29, but we know that the risk is it will wane. We just don’t know if so, and how fast if so. Seems definitely worth looking into.
