The BioNTech/Pfizer data is out—NEJM & FDA report. I understand this wasn't in the design & the long-term protection from single dose is open question. But given this sharp drop after 14 days after one dose, can someone explain why single dose isn't very very high on the agenda?pic.twitter.com/sxqwACGulz

Second dose important for overall durable immunity in animal models. This study tells very little about even medium term efficacy of a single dose, unfortunately

Right, but the alternative is zero vaccination for hundreds of millions—maybe billion—people next year. Why not start with single dose at least for a big enough group so that we know more as soon as possible? The trade-off is very real here.

The reason *might* be that if we (say) give 100M single dose, and it is only 50% effective over 6 months, whereas 50M given 2 doses is 90% effective, you protect similar numbers of people. BUT if you prioritise 2 doses to most vulnerable, you may be saving far more lives.

Yeah. On the other hand, the next three months look terrible.

agree it's about trade-offs. But I would prioritise getting vaccine to most vulnerable. Since distribution and administering is its own bottleneck, the two dose v. one dose won't actually make much difference for next month or maybe more.

Here's another argument. Say the efficacy is closer to 80% in the time scale of a month. If you expect the vast majority of COVID mortality in 2021 to occur in January and February (as I do), it may make sense to vaccinate as many as possible then. After, we will have more doses

Do we know how many doses we should have upon approval from Moderna as well? Seems like if you start adding them up, we may have enough between different vaccines to overcome scarcity of any one vaccine?

In CA, what I've heard is 300k Pfizer and 700k moderna by end of the month..
