    i know i'm a bit late to the party, but to anyone who thinks #SARSCoV2 FCS is engineered, if you were actually engineering in an FCS: 1) you'd use something you've seen before, say RRSRR in HKU1. turns out this particular one *does* show up in some strains of FIPV but it's rare.

        & given how our two most worrying VOCs both mutated that proline away (which abrogates O-glycosylation: https://www.biorxiv.org/content/10.1101/2021.02.05.429982v1.full.pdf …) & specifically to positively-charged AAs that are optimal there... yeah.

        also i will take this moment to remind you just how goddamn quickly you lose that site if you grow the virus in cell culture. like it's an uphill battle keeping it intact. it's only selected for in hosts, outside of them, it goes away *FAST*.

        ...wanted to get an idea on how different FCSes drive both: 1) cleavage rates & 2) pathogenicity in different viruses. in the process realizing that having a proline at P5 is *hilariously* not optimal & this is the sort of thing that's just good enough, i.e. natural insert.

        this thread was written as an afterthought b/c i was just amused at how comically silly of an idea that that site was *engineered* was. there's so many things wrong w/it, starting w/the fact that it's *not* somehow magically better than you'd think, it's indeed resoundingly meh.
