Ok, that is clear but can it be (asking because I don't know) that they are separated? Such as some one that has sex A, gender B but does not consider themselves trans?

No, because I cannot think of a case wherein that would not be misgendering. Sex and gender are both constructs humans have created. If you want to know if somebody has a vagina or penis or neither you can ask about genitals.

Again, the point is to include preset independent variable. sex, gender, can both be presets to be used at the discretion of any given researcher. We don't have to make choices here other than to include them in the list of presets. >
