developing Win32 applications without ever touching any code written by Microsoft is thoroughly strange but entirely possible.
now I'm wondering how hard it'd be to unintentionally end up with a program that runs fine in wine but not windows
you can use linux syscalls under wine (I mean, there's no real reason that would not work)
Also Linux and Windows use different registers for TLS/process-specific info (fs and gs), which is really handy for Wine to be able to work at all (since Windows apps will happily contain code that directly reads such stuff). Wine processes are a weird combination of both.
And then a Linux library that Wine calls makes a thread and calls back into Windows-land callbacks and everything nondeterministically explodes because you have a pthread in a Wine process that isn't a real Windows thread. Yeah, that was a fun Winepulse bug to debug.
