    The target was discovered through unbiased phenotypic screening & extensive validation by and colleagues. They also found that JTE-607 acts as a prodrug. We played a small part by showing inhibition of 3' end cleavage in vitro

    Surprise finding that the CPSF endonuclease that cleaves pre-mRNAs to generate a 3'-end is the target of JTE-607, a compound that inhibits production of proinflammatory cytokines, & promotes cell death of AML/Ewing's sarcoma cell lines

    We also determined a structure of FANCD2 alone and found it is a homodimer, in a closed conformation that cannot bind DNA with occluded monoubiquitination sites. We propose a model for how FANCD2 and FANCI act as a signal to recruit DNA repair factors.
