    Ectopic expression of a transgenic receptor is likely disruptive to natural cell dynamics. For example, the kinetics/balance of recycling receptors on the cell membrane may be changed. An inert receptor would better approximate potential disruptions than a fluorescent protein.

    I often see fluorescent proteins used as controls in chemogenetic/optogenetic experiments, but I think this is flawed. A fluorescent protein does not recapitulate the processing, transport, and subcellular localization that a receptor does.
