Can you briefly explain the significance of these findings for a layperson interested in the technology?
-
Certainly. By 'technology' I assume you mean gene editing, for which Cas12 is used. We see value in turning off gene editing when it is no longer needed (i.e. after the desired change has been made), but when the 'scissors' (Cas9 or Cas12) are still roaming around the nucleus
-
Small protein inhibitors like these, grab onto those molecular scissors, turn them off, and keep them turned off. This is useful for applications in human cells, and perhaps, for preventing gene drives when not desired. Other applications with Cas9 inhibitor AcrIIA4 (2/3)
-
are further developed and there are a few papers where it has been applied to stop Cas9 or dCas9, lowering off target effects, reducing toxicity in stem cells, and blocking gene drive. Now people interested in these applications can do the same with Cas12 (3/3)

Finally, Cpf1 inhibitors are available!
-
Yes they are! Plasmids constructed and tested in Joung lab, available on
@Addgene! https://www.addgene.org/Joseph_Bondy-Denomy/ … -
Cannot wait to test them. Thanks Joe!

I can't keep up with your work Joe ! Congrats for your paper. So what would be next, more unknown anti-CRISPR and/or anti-anti-CRISPR?
-
Thanks Gaetan! As my
@ucsf colleague and mentor@fraser_lab tells me, it isn't beer OR tacos, it is beer AND tacos. I vote 'and'! Keen to uncover new Acr mechanisms also, hoping to find some that teach us something really new about CRISPR-Cas function - 1 more reply
