Estimating the population of monthly twitter users (as defined by: did tweet) from `statuses/sample` is something I'm going to have to do for engineering purposes, apparently. Which is boring but I haven't gotten to do any estimation in a while, so...I want to lol.

I have a huge DB of Twitter users and some other data that should help you. Plus methods of discovering user accounts that are never active, etc.
I'm not sure if this would work for your research, but if we narrow the window of time down, some things would be easier.
Questions like -- "How many user accounts are created in a typical day, etc."
