welp that settles it as far as I'm concerned absurd it took this long for someone to do monkey studies on thishttps://twitter.com/davidrliu/status/1263960572032430080 …

Can hydroxychloroquine prevent #SARSCoV2 infection if taken before exposure? Can it treat early or late #COVID19 infection, with or w/o azithromycin? This study in monkeys (prophylaxis n=5, treatment n=18 total, control n=8) concludes: no, no, no, & no. https://www.researchsquare.com/article/rs-27223/v1 … pic.twitter.com/HXGEJqIN9D

does a study of this size, in another species, provide convincing evidence of anything in your field . . . usually?

primate studies are usually pretty small n, it would be *nice* to have a larger n buthttps://mobile.twitter.com/halvorz/status/1264330825958653954 …

by itself it's not super convincing but in combination with everything else it's enough to push me over the line from skeptical to "nope" prophylaxis is the absolute minimum for an antimicrobial, if you can't even pass that bar you're fucked

if it were claiming a positive effect it would be less convincing, but you can just look at the graphs above nothing, not even a hint

heh my day job is partly yelling at people for running underpowered experiments do they report anything like a minimum detectable effect here? just seems like you might be able to rule out some kind of night-and-day effect here but nothing subtle

we're not really interested in subtle effects here practically speaking, if you can't see anything in a small n animal study, nobody is gonna want to give you more animals, or god forbid go into humans

animal studies almost always look *better* than human studies, so if it starts out looking shitty in animals... hypotheses are abundant and low value gotta triage

would the reason for animal studies being better have anything to do with regression to the mean?

that+much more controlled conditions+(for mice) genetic homogeneity+disease models with more or less relevance to humans like, for infectious diseases inoculum size and timing is the same for every animal, you can track the course of disease very precisely

for cancer and neurodegenerative diseases basically every animal model is artificial in some way but yeah regression to the mean is probably a big factor too
