He's making a huge assumption that all of the sample collection and PCR testing is being done by highly trained and experienced technicians, which it is not, due to the very rapid increase in the volume of testing. He also ignores the possibility of nosocomial infections.

You can't get a false positive by failing to collect enough sample. PCR is qualitative not quantitative.

Yes you can if you run enough cycles. If you can't detect anything until you've got up to 40 replication cycles the chances are you're copying such a miniscule piece of genetic material that it could be anything.

Again, not collecting enough sample is far more likely to produce a false negative in PCR rather than a false positive. I've done PCR for nearly 30 years. If your negative controls work and don't amplify anything, it's incredibly unlikely that too little template will amplify.
