Question for my designer friends: I'm considering making a new prototype and wondering if it's worth making the whole thing IN Tabletop Simulator so it can be edited during the session when necessary. Is there a strong reason not to do this?

I’m not sure what you mean by “make the whole thing” in TTS. Do you mean never printing anything out, and only ever iterating in TTS? Or crafting a game that will only ever be played in TTS?

The first one! Crafting the prototype directly using the tools in TTS so I could tweak it during the game. As for a physical production, that's not a concern at the moment. I just want to see if this concept can work.
