Is it possible for a font to unlock UI building blocks that recreate GUI elements? For example filled rectangles w slightly rounded edges?

All is possible. This is a good idea. Consider there are already rounded edges (U+256D to U+2570) or do you think aren’t enough?

I worry a little bit about how critical it is to get terminal/editor row/column alignment just perfect for creating opaque border shapes.
