    I should add that streptococcus or Staphylococcus are commensal pathogens, hence the immune response against Cas9 effector -> consider Cas9 orthologs or other effector enzymes from non commensal bacteria to circumvent immune response but not sure it would work better

    As I mentioned yesterday, I am more worried for on the immune response to enzyme SpCas9 and SaCas9 rather than off-target effects. I don't know if the use of Cas9 orthologs would circumvent the immune response.

    Agreed -> 'A flawed study shows how little we understand 's effect' Besides off target effects and target sequencing to capture those, antibody response to SpCas9 is another concern, potentially more important than off target effects

    If we want to consider using in AAV vector for Immunosuppressive drugs such as corticoids or adoptive transfer of Treg cells should be considered to circumvent the immune response. For ex-vivo , this doesn't seem to be an issue.
