"Would you change anything about your academic experience?" There is no "right" answer. You should be honest about how you feel about your evolution as a scientist and thinker. If you wouldn't change anything, then don't. Explain why and give examples.

"What are your longterm career goals?" Connect your goals to what the institution or program has to offer for professional development.

"What would you do if you weren't pursuing __( Master's, PhD, MD,PhD, etc.)?" This is a good opportunity to highlight your other passions or hobbies. Be honest. Also, there are many transferrable skills between other fields and STEM.

"How would you follow up with your current or last research project?" Think of some follow up experiments to answer the next question. Suggest alternative experiments if something did not work. Troubleshooting and thinking ahead are valuable traits in graduate school
