Wow tomo is soo cool
... dreaming of the inside of the nucleus

Thanks! Inside the nucleus like this? Or do you want higher mag for those nucleosomes? A lot of groups are going for that, so you won't have to dream long
For now, you can check out the work of @MitosisLab

Interesting how ER-bound ribosomes are arranged in lines. Is this real? Known? Is this mRNA-driven process?
-
Indeed the arrangement of the ribosomes at the ER membrane is intriguing! To me, this would have to do with how Sec translocons are arranged, as RNCs are docked on the membrane or maybe not?

i envy your patience with segmentation!

Segmentation is zen
for me, but I unfortunately don't have much time anymore. Fortunately there's Antonio Martínez-Sánchez's TomoSegmemTV: https://sites.google.com/site/3demimageprocessing/tomosegmemtv …
It did all the heavy lifting with these membranes. And much smoother results than coloring by hand.

this is super cool! wonder if you can make these great images also leaf tissue or roots?
#proteasome rocks! -
That's the plan!

Tissue requires HPF lift-out (see: https://twitter.com/bengeliscious/status/1155878391578529792 …). The large vacuole complicates HPF of plant tissue such as leaves, but I think it can be done. The root tip freezes well and is a very good target for this technique!
