How long do those interactions last and can you measure the strength of them?
-
They last for >30 min but we do not record that long. There is even internalization of some of these beads. We quantified extensively force generation by T cell during activation (Sawicka et al. Mol Biol Cell. 2017, Basu et al. Cell 2016, Husson et al. PLoS One 2011).

And the MTOC will oscillate between the two synapses
-
Hi Heffrey, I wonder what timescale you would be expecting? I'd guess the MTOC would get in the vicinity of the synapse typically 10 min after receptor engagement, but what typical timescale would you expect for these oscillations?

Those of us that have activated T cells with Dynabeads, have seen T cells forming synapses with multiple Dynabeads. And this video is cool because it shows how fast a T cell forms those SMAC complexes!

Given, the high affinity of antibodies, was this study tried with TCR cognate peptide-MHC complexes? If not my postdoc PI, Jim Riley at
@PennMedicine, has the artificial APC protocol that we developed that can test my question.#immunology - Još 3 druga odgovora
