Our 2nd insula paper is out in ! We found that insular cortex represents current bodily state AND uses external cues to “simulate” future changes to bodily physiology – “How do I feel?” + “How will this make me feel?” More in THREAD…1/7

Exciting new research on the insular cortex, neural basis of interoception from labs of & Brad Lowell, first author -- basic research that could one day help us better understand eating disorders, obesity, addiction etc.

New interoception paper hot off the press from Mark Andermann's group : First, this is a super-cool experiment and methodology. The causal power with manipulating neural circuits related to interoception is unparalleled.

Find out about Calcitonin receptor neurons in the NTS! They produce not just satiation but long-term satiety without aversion. Way to go for suppressing feeding without nausea.

We speculate that these cortical “simulations” along different dimensions/axes of population activity serve to guide and prioritize behavior. For example, how will eating salty food when you’re both hungry and thirsty affect energy and fluid balance?…6/7

But what do food/water predicting cues actually mean in insular cortex? We looked at the patterns of activity that these cues evoke and found that they drive a transient “simulation” of a future satiety state that would only occur hours later…5/7

Insular cortex was not “fooled” by the hypothalamus: artificial activation of hypothalamic “hunger neurons” and “thirst neurons” in sated animals restored responses to learned food/water cues. BUT it did NOT affect ongoing activity, which still reflected to true bodily state…4/7

We followed-up on our previous work () and imaged insular cortex population activity in hungry or thirsty mice as they gradually became sated. insular ongoing activity patterns reflected current bodily state, not behavioral state (arousal, motivation)… 3/7

Insula is considered the “primary interoceptive cortex”. It’s hypothesized to integrate visceral-sensory info about current bodily state (hunger, thirst, temperature, etc) with previously learned info to potentially compute interoceptive predictions and prediction errors…2/7
