With respect to choosing IMPORTANT TOPICS - I would advocate that the genetics of mental disorders and cognitive traits rank top on my list. We need to build capacity in brain sciences and we need the best and brightest to join us in this important work. 🇦🇺🇩🇰

CLiP is well powered for highly polygenic GWAS traits, detecting heterogeneity in PGC schizophrenia cohorts (from the 2014 Nature paper) across SNPs totaling 3.4% of variance explained. 6/6

Aside from generating insights into disease etiology, CLiP can also be used to evaluate polygenic risk scores when heterogeneity is undesirable, such as for transferability across demographic groups. 5/6

We also develop extensions of CLiP to handle quantitative predictors (such as gene expression) and quantitative phenotypes. 4/6

CLiP takes advantage of expected correlations between associated SNPs when ascertaining for cases. These correlations are expected of all logistic and liability threshold models, but in heterogeneous cohorts they are altered. 3/6

We developed a novel method called CLiP (Correlated Liability Predictors). CLiP calculates a Z-score testing for heterogeneity in case/control phenotypes. Requirements are only genotype and phenotype data and summary statistics for a single trait. 2/6

Pleased to announce new work posted to biorxiv led by my colleagues and Jie Yuan It is often assumed that complex disorder (like schizophrenia) are heterogeneous. Here, we utilize GWAS data to identify heterogeneity within a complex phenotype. 1/6
