Be aware of using AAVs to deliver shRNAs though https://www.nature.com/articles/nature04791 … https://www.ncbi.nlm.nih.gov/pmc/articles/PMC2929739/ … https://www.ncbi.nlm.nih.gov/pmc/articles/PMC2841193/ …https://www.ncbi.nlm.nih.gov/pmc/articles/PMC3747597/ …

This is such an important question! Best advice I can offer is to get any subsequent preps (for the same project) made in the same facility. Every facility users different titring methods so it's soooooo difficult to compare doses from batches made in different places.

Thank you!

AAV is a "you get what you pay for" product. Don't look for cheap, look for quality. Cheap vector --> variable, full of contaminants (both DNA + protein), low potency, empty capsids, immunogenic. Vendors need to give a CoA and know what they're doing (UPenn core, SignaGen, etc)
-
I heard about the UPenn core from a colleague. I'll contact them as well :)
