Assuming they are in the US, yes. If they aren't, lacking a country that recognizes universal copyright law (such as China) there's not much that can be done except by YouTube, and they'll only get involved after being told to by the courts.
Here in France sending a C&D letter to YT makes then usually react quite fast.
I believe the EU has laws in place that mandate they do, and especially after Google got hit with a billion dollar fine a year or so ago they take it more seriously.
