It’s a good idea to invite post docs to sit in on candidate chalk talks. A great opportunity to learn and prepare for their own job searches.

What happens to transcription factors during DNA replication? Check out this beautiful work by and to find out!

6/6 We conclude that chromatin environment during S-phase allows binding of transcription factors to replicated DNA, but preclude recruitment of RNA polymerase II, thus resulting in transcriptional inhibition.

5/6 Following this transient eviction, abundances of all three TFs increase in replicated regions, to an extent that is (at least) proportional to the increase in DNA content.

4/6 By contrast, we observe a clear transient depletion of the three TFs from regions that are being replicated, suggesting eviction of these factors by the progressing fork.

3/6 We find that RNAPII binding-pattern remained largely unaffected by replication. First, we could not detect signs of its eviction by the progressing replication fork. Second, also after fork progression, its binding pattern remained largely insensitive to the change in dosage.

2/6 We followed budding yeast progressing synchronously through S phase, and defined at high time resolution the DNA binding patterns of RNA polymerase II (RNAPII) and of three specific TFs regulating a large number of targets (Reb1, Abf1 and Rap1).

1/6 We asked whether the replication-specific chromatin environment limits accessibility of regulatory factors to DNA.
