1/2 Adjustment for covariates in the Cox model improves power and increases the likelihood of PH holding. You were caught by the "ill-defined hazard ratio without adjustment" problem. See ANCOVA chapter in

Most angiomyolipoma can be monitored without any active treatment. The 4cm cutoff for intervention is not valid. Bleeding risk is only 2%. Nephron-sparing surgery or embolization are the preferred treatment options ⁦⁩ ⁦⁦

Lord’s paradox is discussed here: and here . Basically, analysis of change scores is almost certainly wrong. ANCOVA is right if certain (implicit) assumptions hold.

This plot from Statistical Issues in Drug Development chapter 7 shows relative asymptotic variances of change score and ANCOVA compared to not using the baseline at all (just raw outcomes) as a function of the correlation between baseline and outcome.

One of the nice things about Andrew Gelman's blog is how benign posts routinely produce deep debates about foundations of statistics. This time, the concept of "calibration" (see comments): My attitude (new box in 2nd ed of my book):

This plot, from 's book SIDD shows how the RD and RR change as a function of the baseline risk in a compactor group, yet for all the various RD and RRs, the log odds (and thus OR) are the same.

This is a major milestone of progress in cellular therapy in lymphoma! And it was initiated in the MD Anderson Lymphoma Moonshot Project. I am so glad to be a co- author.

3/2: we do not observe CfB: we compute it; and in using CfB as an outcome information is lost. That information is better preserved in mRcB bc you can conditionally express the observed and expected objective of the action of interest—the ‘do(x)’.

No. Example: semiparametric models are the preferred method because of transformation invariance resulting from treating Y as ordinal. Difference in two ordinal variables is no longer ordinal in general. Think pain severity scores.

1/2: Consideration of intent and utility (psychology)may be illuminating for the issue of change-from-baseline (CfB) vs models for response conditional on baseline (mRcB). Humans intuitively construe effect as systematic change. But this intuition may make mischief for analysis.

Unfortunately this document contains several statistical misconceptions and will perpetuate invalid responder analysis using change from baseline
