    We believe that cells which have smaller nuclei (like Dictys) will have a mechanosensitive module driven by mechanosensitive ion channel on plasma membrane.

    Piezo null mutants chemotaxed very poorly under mechanically resistive environment. Blebbing is necessary to chemotax under such conditions as has been shown earlier by Kay lab.

    But, amongst all the mechanosensitive ion channel mutants, Piezo null mutants did not show protrusion switching under compressive loading.Pzo null mutants are expressing LifeAct-GFP while WT cells express LifeAct-RFP. WT cells formed blebs when load was applied but not mutants.

    Protrusion switching occured at a very fast timescale(~20sec). In this movie, Dictys are expressing a marker for F-actin and a uniaxial load is applied from top using a cell squasher system.

    It is to no surprise that academic research has evolved from one of the most fulfilling careers to more stressful one especially for ECRs. Adding to this is the non-permanency of the role and fact that most people immigrate to places far away from loved ones and families.
