I don’t think so, although maybe I’m missing something? If you hang random junk off a nucleoside, it’s pretty likely to screw up the polymerase.

That one's easy, it's the 1′-cyano on the ribose: "The presence of the 1′-CN modification in 4 was found to be critical in providing selectivity toward viral polymerases and avoiding the significant toxicity (CC 50 < 0.01−0.15μM) associated with the unmodified C-nucleoside 7."
