I don’t think so, although maybe I’m missing something? If you hang random junk off a nucleoside, it’s pretty likely to screw up the polymerase.

The iterative optimization steps would be to get the IC50 down, make sure it gets to where it needs to go (not too lipophilic etc), and selective for virus/human polymerase. Pretty standard medchem analoguing Iexpect
