In-situ sequencing (of DNA/RNA and DNA-tagged antibodies, at nanoscale resolution w/ ExM) could become the new new microscope... What else could become the new new microscope? Protein sequencing? Imaging inside scattering media? Electron diffraction? DNA ticker tape recording?

Great post! I liked how it was concluded, especially how you describe sequencing as the new microscope and biologists as the new engineers. As we understand more biology, we can use tools to actively shape it!
-
I guess the next question to ask would be: what do you think is the next "new microscope"?

There are still a ton of new tools emerging from physics: optical tweezers, electrode arrays for neural recordings, acoustic liquid handling, etc.

Another cool example just posted today:https://phys.org/news/2018-11-high-resolution-atomic-movie-photosynthesis-date.html …

We might have talked about this already, but "Image and Logic", from the father of tool-based scientific revolutions Peter Galison, is absolutely worth reading. The book charts tool-based revolutions in physics and astronomy. https://www.amazon.com/Image-Logic-Material-Culture-Microphysics/dp/0226279170 …

It's plausible that a significant proportion of the advances in biology, especially regarding genetics and diagnostics, will be driven by better statistical and machine learning tools.
-
I'd distinguish here between advances in biology and advances in medicine/bioengineering. Unless you can reverse engineer a trained model to understand how features interact, you're not really learning the underlying biology.

Really interesting post on tooling & compounding effects. What're some good reads/books on biology history to extrapolate these insights?
