    Starting sabbatical in Bay Area. Last sabbatical I ambitiously began writing a book on economics and finance for the general reader . Ouch. So this time my only ambition is to hang out, talk to new people and see what happens, preferably within biology.

    Protein abundance should affect selection, not sample preparation errors, so this is evidence that the higher error rate in E. coli than S. cerevisiae is real. and I predicted this countintuitive result in 2011 from drift barrier theory.

    Curves like this have the potentially serious problem of making errors/reads a function of a correlate of reads. We directly a Poisson error function, handling both this problem and heteroscedasticity. Binned data are to show linear shape only.

    S. cerevisiae has a much lower error rate, and no relationship - except for the C->U errors, which were measured with a better protocol, and are at high enough rates for selection to care.

    The E. coli relationship was steeper for G->A mistranscription events. It was non-existent for C->U because too much deamination happened during sample prep rather than biologically.

    Our latest paper documents patterns in transcriptional error rates. When they are high, there is gene-level selection to reduce them in highly expressed genes. When they are low, they are equal across genes.

    We reconstructed regulatory networks for yeast promoters in this way, and found that weak regulatory interactions explain the vast majority of expression. These two promoters encode the same expression level, but using very different regulatory strategies!

    Most yeast TFs (and human too), have low information content and so are predicted to occur often by chance in random DNA. How often? The average random 80-mer has about 138 binding sites for 68 different TFs (!)
