Types of peer review: 1) 3 anonymous scientists write secret reviews in a process that takes a few months 2) dozens of known experts write comments that anyone in the world can see hours after a paper is posted (e.g this thread 👇) Explain to me again why is 1 >> 2?

Again and again, reporters ask me: "Why all the vitriol? It's just a work of fiction." Ah, privilege. What a drug. If you didn't grow up Mexican American in a Mexican American community, you didn't spend your childhood hearing about (& experiencing) the harm done to us. 7/

Yup, language matters: "words carry our assumptions and project stigma and shame" Important to consider today on and going forward.

As time goes by, learning new things might get harder, but not impossible. Keep on learning! And stay tuned for future updates! Grateful for the support from who made the study possible! 10/10

I believe this approach can interestingly be generalized to other studies on perceptual learning (e.g. visual, tactile, etc) & be extended to other modalities (motor learning, memory...). 9/

Our study is the first to use PET scans in rodents to demonstrate how perceptual learning changes cholinergic function in a region-specific manner. For more details and pretty figures, check out our full paper 8/

Learning also resulted in microscopic changes in auditory cortex (Figure 4 in the paper)

With this study, we have shown that: 1⃣ intensive training can lead to significant learning even in the very old brain 🧠 2⃣ auditory training resulted in increased cholinergic function specifically in the brain area that processes sounds 🔉 7/

After training, we found an increase in cholinergic activity specifically in the part of the brain that deals with hearing 👂 but not in other brain areas. We concluded that yes, the cholinergic system itself changed with training. 6/

'Au' is the auditory cortex, a part of the brain that identifies sounds. The scan shows more activity in this part of the brain after training to differentiate between sounds

We also scanned their 🧠 before/after training to map changes in their cholinergic system. We used an imaging test called PET scan. On these scans, the brain areas with greater cholinergic activity showed up as bright spots. What did we find, you ask? 6/

How to analyze PET scans (Figure 2 in the paper)

We trained old rats🐭 to identify different sounds 🎶 Each time they identified the 'target sound' they got a chocolate pellet 🍫. If they got it wrong, they just had to wait for a few seconds for their next chance. Attention? ☑️ Motivation? ☑️ 5/

They worked really hard, training five days a week for ~9 weeks 🗓️

Also, there was another question that kept bugging us: Tons of studies have shown that activation of the cholinergic system remodels the brain, but does the cholinergic system 𝑖𝑡𝑠𝑒𝑙𝑓 change with learning? Here's what we did to address these questions... 4/

Perceptual learning is facilitated by the 𝐜𝐡𝐨𝐥𝐢𝐧𝐞𝐫𝐠𝐢𝐜 𝐬𝐲𝐬𝐭𝐞𝐦, an ensemble of brain cells that enhance attention & motivation. But the cholinergic system degenerates w/ age, making learning📔 harder as we grow older 👵 Can we help older brains learn better? 3/

W/ practice, we can become better at distinguishing between diffrent sounds🔉, images🖼️... This process, called 𝐩𝐞𝐫𝐜𝐞𝐩𝐭𝐮𝐚𝐥 𝐥𝐞𝐚𝐫𝐧𝐢𝐧𝐠🧠, supports language acquisition, musical expertise, and even professional judgments, such as the interpretation of X-rays🦴 2/

Nice to see a new preprint from and colleagues re for response 🙌. Polygenic risk scores do not predict antidepressant response . However patients with high genetic risk for schizophrenia more likely to be non-respondents
