Question for all the smart people who follow me: can you explain simply why the J&J vaccine only needs one dose but Pfizer/AZ/Moderna etc need 2?

I don't think there is any reason in principle why the other vaccines couldn't work just as well as JJ in one dose, but would be love to hear from a true expert.

The extremely limited data we do have, with many many caveats, in fact suggests the mRNA with a single dose works better than J&J. But many variables differ.

I'd defer to an immunologist on this, but my understanding is the expectation for an adenoviral vector is a robust immune response from a single dose, because that's what we typically see with adenoviral vectors.

Plus, the potential for an immune response to the vector itself can make expression after the second dose less efficient and more likely to produce side effects

J&J trial results 66% effective overall at protecting against COVID-19 but prevented 100% of virus-related hospitalizations and deaths. Not an issue of mechanism, I don't think, but of measured outcomes. It's likely that >1 dose will prevent more cases, no?
