Does only four fingers make certain ASL symbols hard to do?
Not yet. The game doesn't have lengthy exposition delivered by her. Just puzzle hints, reactions and emotional responses. Simple gestures.

There are very few signs that require all five fingers beyond numbers. Sign is adaptative too, so Quill can be easily understood still.
