This strongly resonated with me my entire career for a few reasons. A lauded scientist made time for a kid from overseas, and was self-deprecating, generous and funny. I imagine I’m not the only person that Phil Leder inspired. Thanks Dr. Leder. RIP. 8/8

“You’re going to help with an experiment” he said, and put the plate up against the side of my leg, himself and the trainee now both clearly seeing the plaques. He then thanked me for my contribution to his lab’s work. 7/

One of his trainees comes up to him with a plate that I subsequently came to recognise as showing phage lysis, and shows it to Leder. He squints at it, not clearly able to see the little holes. Then he looks at my black trousers. 6/

I asked him about how he came to invent his new approaches for ‘recombinant DNA’ cloning techniques that gave him insights into the molecular biology of the immune response. His answer “Because I’m lazy. The usual approaches were too difficult” 5/

I arranged a time slot to visit and put on the most respectable clothes I had with me, including a pair of black trousers. Phil Leder comes out of his officer to greet me, and we go for a all around the lab. 4/

I only knew of high-profile scientists, so that’s who I contacted. Only one responded to this innocent Irish lad. Phil Leder. He gave me the number of his assistant and said I could come by the lab any time. 3/

I was a teenager over in Boston on a J-1 visa, working as a busboy and a security guard, whatever pulled in a bit of cash so myself and could have a bit of fun. Before going, I posted letters to prominent Boston scientists asking whether I could come visit. 2/

Today is the anniversary of the plane crash that killed Buddy Holly, Richie Valens and The Big Bopper in a field in Iowa. The Big Bopper, suffering from flu, asked Holly's bass player Waylon Jennings for his seat on the plane. Chantilly Lace:

Respect to the first person who figures out why this is the only crown that matters...

A crown. Looks a bit cheap. Why the big deal?

For I’ll start by reading these two books. My old paperback copy of the Malcolm X autobiography fell apart years ago, long overdue a rediscovery on my part. The Maroon Societies book looks very academic, hoping for some good stories embedded within.

Emily Hodges has been doing great work on DNA methylation since her days at . I always liked this 2011 work on the expansion and contraction of hypomethylated regions during haematopoietic differentiation:

If we have the answers, the question becomes how they are enforced. Probably by making them requirements of funding agencies and journals.

First the chromatin opens, then the DNA methylation is lost. The chromatin opens because transcription factors (TFs) target those loci. TFs tell DNA methylation and nucleosomes what to do at cis-regulatory loci. Not the other way around. Your textbook needs a rewrite.

Amen x 10. Adding an eleventh (or addendum to number 7) — negative results of rigourous research should be published and celebrated.
