Chromosomes can't be changed. Anything else that is associated to a certain sex or gender can be changed, some parts take a lot of effort, but they can be changed.
Which is not base for anything cause 99% of people never tested their chromosomes. Also, there are no single-sex spaces. No law defining those. Gendered spaces can be created by the facility provider if they think it's reasonable. Trans woman can use woman spaces for decades.

Chromosomal sex is not be needed to be tested in the vast majority of people. Sex is clear in the vast majority of people also. Those in which it is not clear are not some third sex, they still have a sex which is either male or female.
