Thanks to for the nice news article on our recent study showing how loss of Tsc1 from dopamine neurons reduces their output and impairs behavioral flexibility. Study led by

Tuberous sclerosis complex is characterized by tumors in multiple organs including the brain, lungs, kidney, and skin. Learn from how human stem cell modeling of neurons and glia, and TSC1 and TSC2 genes, impact neural development and function.

The Hostage Brain by McEwen and Schmeck was very influential in my life and career. I still have my copy bought over 20 years ago...

I discovered Bruce McEwen's research as a sophomore in college and it was the reason I wanted to become a neuroscientist. I was lucky to work with him briefly in graduate school. He was a profoundly kind and thoughtful person.

(4/4) Overall, we conclude that alterations in the striatal direct pathway may be a previously unappreciated driver for ASD-related behaviors (including restricted and repetitive behaviors) in individuals with TSC

(3/4) Behaviorally, this results in enhanced motor routine learning on the rotarod, a phenotype that has been observed in multiple ASD mouse models (including Nlgn3 , Cdh8 , and Cntnap2 )

(2/4) Mechanistically, we find that this is likely due to a loss of endocannabinoid-dependent long-term depression (eCB-LTD) at cortical-dSPN synapses.

Main Takeaways: (1/4) In this study we show that selective deletion of Tsc1 (which is mutated in the neurodevelopmental disorder TSC) from direct pathway striatal neurons (dSPNs) has profound effects on their synaptic physiology, causing increased corticostriatal synaptic drive.
