@AdamMarblestone In the ExSeq paper, the passivation step, did the team see it coming from the begining based on what you know about the chemistry of the ExM reagents? Or was more of a "surprise"

Was a surprise — we thought just adding more magnesium (to cancel the first gel charge) after embedding in a second gel for stability would work, but it didn’t. Was also unclear if passivation would work.

By 'more' magnesium you mean more than it is used in some other step? I can't see uses of Mg in the ExM or FISSEQ protocols, or in the paper
