Evolution of rhizobial lineages was replayed in an evolution experiment where plant pathogen Ralstonia solanacearum evolved the capacity to nodulate and invade Mimosa pudica. The improved infection required the Phc quorum sensing system.

Colistin AST w/ disk diffusion and gradient strips performs poorly. Previous results showed that use of calcium-enhanced agar improved results with mcr-1 isolates. Is this method appropriate for use with non-mcr-1 isolates? Nope.

Hemagglutinin destabilization decreased virulence of H1N1 influenza virus in mice by boosting early infection in DCs, resulting in the greater activation of antiviral responses, including the type I IFN response

Studies suggest a high genetic barrier to resistance, but it's an important aspect to consider in moving to human treatment. Tim: “I don’t think we can ever stop developing antiviral drugs...you really need to be thinking 2 or 3 steps ahead.”

Remdesivir has already been studied in people: “If you have an experimental drug treatment, you can do experiments in cell culture or in mice, but you still don’t know if it’s safe in people. Remdesiver has already gone through Phase I, so we already know it’s safe in people."

Tim has studied Remdesivir in mice, a close but not identical animal model of disease. CoV disease in people takes place over a course of about 2 weeks. In mice, the disease is similar, but progression is faster, taking about 1 week.

Remdesivir works against every coronavirus tested so far, including viruses with the most divergent RDRP sequences (nsp12 below). The gene for RDRP is more highly conserved than the gene for the spike glycoprotein

One drug Tim studies is remdesivir (RDV), a nucleoside analog that inhibits the coronavirus RNA-dependent RNA polymerase (RDRP). Note: Tim has not looked at RDV inhibition of 2019-nCoV, but similarity of 2019-nCoV to other coronaviruses makes RDV a promising inhibitor

SARS-CoV prefers ciliated epithelial cells and uses ACE2 as its receptor. MERS-CoV prefers nonciliated epithelial cells and uses DPP4 as its receptor. [2019-nCoV appears to use ACE2 as its receptor: ]

In H. pylori, the 3 Cag ATPases have nonredundant functions required for CagA delivery. The energetic requirements for phenotypes associated with intracellular entry of nonprotein constituents differ from that for translocation of CagA.
